Interactions of Soluble Recombinant Integrin avb5 with Human Adenoviruses
نویسندگان
چکیده
av integrins have been identified as coreceptors for adenovirus (Ad) internalization; however, direct interactions of these molecules with Ad have not been demonstrated. We report here the expression of soluble integrin avb5, which retains the ability to recognize the Ad penton base as well as vitronectin, an Arg Gly Asp (RGD)-containing extracellular matrix protein. Soluble integrin avb5 reacted with seven different Ad serotypes (subgroups A to E) in solid-phase binding assays. The soluble integrin exhibited different levels of binding to each Ad serotype; however, binding to multiple Ad types required the presence of divalent metal cations and was inhibited by a synthetic RGD peptide, indicating that RGD and cation-binding sequences regulate Ad interactions with avb5. Incubation of Ad particles with soluble avb5 integrin also inhibited subsequent Ad internalization into epithelial cells as well as virus attachment to monocytic cells. These findings suggest that soluble av integrins or antagonists of these coreceptors could be used to limit infection by multiple Ad types. The generation of soluble av integrins should also permit further detailed kinetic and structural analysis of Ad interactions with its coreceptors.

Structure of Adenovirus Complexed with Its Internalization Receptor, avb5 Integrin
The three-dimensional structure of soluble recombinant integrin avb5 bound to human adenovirus types 2 and 12 (Ad2 and -12) has been determined at ;21-Å resolution by cryoelectron microscopy (cryo-EM). The avb5 integrin is known to promote Ad cell entry. Cryo-EM has shown that the integrin-binding RGD (ArgGly-Asp) protrusion of the Ad2 penton base protein is highly mobile (P. L. Stewart, C. Y. ...
متن کاملIntegrin a5b1-Mediated Adenovirus Infection Is Enhanced by the Integrin-Activating Antibody TS2/16
Adenovirus internalization generally has been accepted to involve an interaction of the adenoviral penton base protein with avb3 and avb5 cell surface integrins. In this study we show that exposure of a panel of melanoma cells to the b1-activating antibody TS2/16 rendered such cells more susceptible to adenovirus infection. This increase in adenoviral infectivity paralleled effects on cell adhe...

Knowing that adenoviral vectors could initiate innate immunity, the ability of E1-deleted recombinant adenovirus (Ad-E1Δ) in induction of B7.1 and IL-2 molecules was studied. Methods: The expression of green fluorescent protein in C1498 cells following transfection of these cells with adenovirus green fluorescent protein vector confirmed the ability of adenovirus vectors in infecting the cells ...
متن کاملCo-expression of recombinant human nerve growth factor with trigger factor chaperone in E. coli
Nerve growth factor (NGF) is a neurotrophic factor that is functional in the survival, maintenance and differentiation of nervous system cells. This protein has three subunits, of which the beta subunit has the main activity. Its structure consists of a cysteine knot motif made up of beta strands linked by disulfide bonds. It can be used as a therapeutic agent in the treatment of many diseases....
متن کاملTumor Necrosis Factor α Regulatesα vβ5 Integrin Expression by Osteoclast Precursors in Vitro and in Vivo1.
Early osteoclast precursors, in the form of murine bone marrow macrophages (BMMs), while expressing no detectable avb3 integrin, contain abundant avb5 and attach to matrix in an av integrin-dependent manner. Furthermore, avb5 expression by osteoclast precursors progressively falls as they assume the resorptive phenotype. We find the osteoclastogenic agent, tumor necrosis factor-a, (TNF) down-re...
